In the mirror universe created by Nero's temporal incursion, Nyota Uhura was a Terran Starfleet officer who served as communications officer aboard the ISS Enterprise in the 2250s. (TOS comic: "Mirrored, Part 1")

After Spock oversaw the defeat of the Klingon Empire on Qo'noS and learned that Commander James T. Kirk had gone to Rura Penthe, he confessed his concerns and lack of trust for Kirk to Uhura. She intimated that he was right to not trust Kirk and that Spock, being a hero of the Terran Empire, would be rewarded with anything he desired. Spock's response that she was all he desire was met with questions of something more he could obtain. He responded that those things were human ambitions. In a move that seemed to surprise Spock, Uhura joined the human crew members that had beamed aboard the Narada with Kirk, informing Spock that Kirk had more to offer. Just before the Enterprise was to be destroyed, Uhura made a suggestion to Kirk that maybe Spock would be more valuable to them as a prisoner, to which Kirk offered to beam her back to the doomed Enterprise with Spock. (TOS comic: "Mirrored, Part 1")

According to Chekov, Uhura translated the Romulan data that allowed them to ascertain that the anomaly is a kind of wormhole. Kirk made the decision to travel to the wormhole and captured the Jellyfish and Spock Prime. It seemed that Kirk trusted Uhura as he left her to watch Spock Prime while Kirk and Sulu went back to the bridge. When Kirk ordered the use of the red matter to destroy Vulcan, he went down to the planet, at which time Uhura took command of the bridge from Scotty by killing him and stopped the attack on Vulcan. It is then that she orders Kirk to call her Captain. It is discovered that Spock was not killed aboard the Enterprise because Uhura beamed him aboard the Narada. (TOS comic: "Mirrored, Part 2")
